报告摘要:

Isoprenoids, the most structurally and functionally diverse group of metabolites with more than 50,000 members in nature, are constructed from two precursors, isopentenyl diphosphate (IPP) and its isomer dimethylallyl diphosphate (DMAPP).  In prokaryotes and plants, these two isoprenoid precursors are supplied by the deoxyxylulose phosphate (DXP) pathway.  The last two steps of the DXP pathway are iron-sulfur cluster catalyzed reductive dehydration reactions.  Recently, using [2-13C, 18O]-MecPP (MEcPP-1), we detected the positional isotopic exchange for the bridging oxygen in MEcPP in presence of IspG without reductants (Scheme 1). In addition, the 13C labelled OH-IPP was converted to the sole product IPP with the retaining isotopic position in the IspH catalyzed reaction (Scheme 2).
